Yes. In sequence 02, there is a mission which has "Get to the fort without being detected" as the main objective when you begin. You are required to give the signal for your men to silence a dog by killing it in order to progress in the main story of the game. Later in the game there is an optional mission to put down rabid dogs.
Yes. There are many dead animals in this game.
1: On the ship near the beginning of the game there are many large cuts of meat hanging up below deck, and there are some live, unharmed animals in a pen nearby. No whole dead animals are seen here, but the area may be upsetting to some.
2: A man is seen holding a dead animal that he caught.
3: At a farm in one of the homestead missions you can see 2 dead pigs, 2 dead turkeys, 2 dead chickens, and possibly others. There is blood in some areas.
4: Dead animals are seen hanging up at various campsites
5: Oliver the innkeeper can be seen chopping up a dead pig outside the inn, and later Warren is chopping up a chicken.
6: If you get near a certain liberation mission in New York, you can see several dead/dying sheep where a rabid dog has obviously attacked.
7: Near the start of sequence 10 you come across a patriot soldier kneeling over his dead horse. This is not optional.
8: In the Tyranny of King Washington DLC there are dead horses and cows everywhere.
There is a sequence featuring a pregnant woman. One of the homestead missions involves getting a doctor to help in childbirth, after which the resident has a baby.

1: You are required to kill wolves that attack you.
2: In some areas during the wolf attack sequence you can see wolves kill a rabbit and a deer in the forest.
3: You are required to kill two rabbits and a deer, as well as 5 additional animals of your choice in order to progress in the game. The skinned carcasses are clearly visible afterwards.
4: Wild animals sometimes attack you when exploring away from the towns. You have to kill them or they will kill you. Sometimes you can avoid them but other times they seem unavoidable. (If you are concerned about this, try to travel up in the trees or on horseback as much as possible to avoid interactions with wild animals)
5: In one of the homestead missions you are required to fight and kill a bear.

7: In one of the homestead missions you go into a mine to lure out a cougar who is shot by another character. The carcass can be seen after.

Sequence 2, The Surgeon: A character is threatened with tortured for refusing to pay protection money. It does not progress beyond the character being hit a few times.
1: There is an underground cave somewhere in the frontier, I think, which is extremely dark, almost pitch black in one area, and it is hard to tell where you are or how to get out. 2: The New York and Boston underground are dark, tight, ans maze-like. 3: In the first Achilles Manor Mysteries homestead mission you end up in a dark collapsing cave with many tight spots. 4: In the Oak Island mission you end up in another dark collapsing cave with many tight spots. 5: In the "The Ghost Ship" mission, you are inside of a ship which starts breaking up and sinking, it's a race to get out and could feel claustrophobic.
This game being set in the mid-to-late 1700s, slavery is still a thing.
