It's complicated, but no. The plot is that an evil carnival grants people's wishes with a twist. There is a bearded man who is obsessed with women who is turned into a bearded lady. He is one of many and isn't really commented on specifically and is not the butt of any jokes. It's also unclear what this transformation is supposed to mean, as he is just wearing a dress and makeup. I'm trans and didn't have a problem with it.

It's a little unclear. The father seems to be very haunted by the traumatic event of not being able to save his son from drowning himself, but it's hard to tell how much of that is trauma and how much of that is feeling emasculated which is how he presents it.

When the boys go in the caravan when the carnival first arrives, there are webs everywhere and a very big spider touches one boy's hand. A woman is wearing a veil in a web pattern and it pans down to her stroking a big, big spider.
Later, when the green mist hits the lightning rod, a lot of spiders come through the ceiling and they are all around the bedroom. Lasts about 2 or 3 minutes
