No, but you will have to fight a friendly but monstrous dog-like enemy multiple times. He is being tormented by spirits, forcing him to fight against you despite otherwise being friendly. When you defeat him you essentially knock some sense into him and he seems unharmed, and the narration explains that from the dog's perspective, he may have just been playing. There are also a number of werewolf-like enemies but they aren't rendered in great detail.
A monstrous household pet is left to wander a certain part of the realm after his owners are captured. The owners don't abandon him, but their captors do, weaponizing him and leaving him to fend for himself.
In a cutscene after you die after winning to the final boss, you temporarily play as Hades during the cutscene. Hades has to abandon his daughter and hands her over to Hecate so he can be assured she can be safe while he deals with Boss 4. As a result she grows up without her family.
the games main antagonist Chronos is Melinoë's grandpa who kidnaped her dad(his son), mom(unrelated) and brother(his grandkid). you do have to fight him. also Melinoë is forced to fight a mindcontroled Cerberus (who is basicly her family's dog) multiple times. but i don't think that counts as domestic violence

Frinos the frog as well as your other animal familiars that you can bring with you on the run cannot be hurt due to a special protection spell. You can also pet frinos as many times as you want.
Arachne is in the game. her profile is very cutesy and cartoonish but you still see her legs and body, she is just a straight up spider. her npc model, outside the portrait, is triggering but very small and easy to cover. overall, as someone with severe arachnophobia, i’d say she’s triggering but not instant panic attack inducing.

Yes, in the surface route your family is afflicted by a curse that means you can’t go up until the surface, so you’ll slowly choke to death for your first runs in the surface up until you find a cure in the cauldron.
Technically due to time stopping, characters are unable to die. However to get to the final bosses of either the surface or the hell route you do need to kill enemies.

Reality can be found extra unstable by taking chaos gates which will transport you to Chaos, a layer outside of time and reality.

the "Price of Midas" item can sometimes be "bought" from a Well of Charon for some of your heath and gives you money in return. the item resembles a dagger that is driping gold as if it was blood. the player is not shown useing the the knife but there is a chance for Melinoë to say "Ow!.. Gold." when "buying" it
