It isn't necessarily portrayed as abuse, but Bela Lugosi was an alcoholic and in addition to being a morphine addict and he does drink in the movie although it is not presented as problematic.

Sort of? A character talks about wanting to go to Mexico to do hormone replacement therapy and have his organs removed so he can become a woman, but it ends up not happening and he apparently does not pursue it again. It’s not a major plot point.

In the closing epilogs for the characters, the epilog for Bunny, who was a trans woman who was never able to get her gender confirmation surgery, is referred to as a "he" twice and is mocked for not having been able to get the surgery.

Ed likes to dress up in angora sweaters to comfort himself. This isn’t presented in a mocking way but some of the other characters find it odd or are rude about it

a character's partner is briefly mentioned having died off-screen. side note: there is some homophobic language used, generally as a joke or a reference to the time period, all of the main characters seem to be very accepting of it however.

Quite a few instances of characters asking Trans/drag characters what's "wrong" with them, yelling at then, being generally angry/hateful towards them etc

Main characters are made to become baptized in the Baptist church in order to make a movie as funded by said church.
There are characters who want to make movies about the 12 Apostles. They end up funding a different movie in order to make money for the other movies and object on religious grounds to some of the content in the film made instead

I dont believe any characters explicitly have s*x on screen, however there is a fair amount of references to it. also side note: there are a large quantity of F-words and other swears, including slight homophobic slurs.

Depends how you define it. The main character is not seen in the movie to achieve the success he hoped for despite constant toil. In the end, he is said to have died an alcoholic and did not achieve fame until after death. Some of the other characters in the movie have happy endings (others don’t), again as mentioned at the end of the movie, but it is not seen.
