Abandonment
0
Multiple people are entirely or essentially abandoned by their parents. This is a non exhaustive list but in example, one character’s father is stated directly to have “walked out,” on his family due to the abusive behavior of his (ex-)wife. Another character’s parents essentially do not raise their child and he has mostly been left to his own devices.

S2 E9 Centers around a main character’s abusive mother. It doesn’t shy away from showing a controlling, mean mother so beware.

The entire reason Nagisa is capable of reading people so easily with a single glance is because he had to in order to survive his mentally unhinged mother Hiromi, who can go from zero to psycho in the blink of an eye. She also constantly tells him she wishes he was born a girl and makes him grow his hair long, despite the fact that he hates it.

-An octopus is dead with a knife lodged in its head, it’s not gory but it’s slightly disturbing.

-An already dead fish is cut into with a knife, although it switches to a different frame before it’s shown fully going in, and no blood or gore is shown. The fish also isn’t real and is in someone’s imagination.

-Gigantic alligator-like creatures are shown dead with slashes in them, however the slashes are just red lines and the alligators aren’t real (they’re the embodiment of tough math problems)

A new female teacher is brought in with a new plot to kill Koro-sensei, her whole character is about seducing people, she fails and then is subjected to "stuff" from Koro-sensei but it is not shown on screen but there is suggestive audio playing. After the ordeal, it's played for jokes and that he was "massaging her". Very uncomfortable for a show about middle schoolers.

What I’m thinking of is more of a science lab but a character is experimented on with no regard for their well being, this could be seen as reminiscent of medical trauma.
There is also a more standard hospital scene when a character is injured and can be seen resting in the hospital for a few days after the injury.

[SPOILERS] The main character's mother always wanted a daughter instead of a son, and this pressure weighs heavy on him. He is forced by his mother to wear long hair and appear feminine. This is generally played for laughs, and jokes regarding gender reassignment surgery are occasionally made at this character's expense. After the students become aware of his home situation, he is apologized to.

The main villain of the first season is far from mentally well. He is a veteran who creates an elaborate plan to use his military training to trap, beat up, emotionally torture and kill a literal child who beat him in a challenge a couple months prior. Also while it’s never really stated or explained, it’s implied Karma has some level of sociopathy

Karma jumps off a cliff in his debut episode, intending to either shoot Koro dead when he tries to save him or die and destroy his reputation as a teacher. Karma is saved in a way where neither occurs and Karma didn’t seem to serious about it, knowing that he wouldn’t have died doing this one way or another anyway.

A character directly comments on how another characters “first line in the series” is a sex joke. At the end of the finale of the first season, a character directly comments while staring at the camera that they are wearing school uniforms because the animators didn’t want to bother with designing and animating new outfits for the entire cast just for the last scene of the finale. Another character follows this up with “who are you talking to?”

The main character is mistaken for a girl multiple times and often teased by his friends for his appearance being more feminine then masculine. At a couple points, he is forced to disguise himself and act like a girl. This behaviour is stopped once they discover how his appearance was brought about by his relationship with his mother and is apologised to. -The teacher is made fun of at the beginning for being obsessed with cleaning, some could see this as ableist to people with Contamination OCD, although the character most likely doesn’t have it.

-The children in E Class are looked down on and called stupid by other students and teachers.

-A charatcer is called a psycho.

-A character seeming to have antisocial personality disorder (sociopathy) is portrayed as violent (however, unless you believe he has APD, it’s not technically ableist).

One of the teachers is an assassin seductress. Many sexually-charged jokes involving her are made.

This anime is very philosophical and, given that a main character is conscious of his own very limited life span, death is discussed in later parts of the series.

Yes, however it’s very minor. There’s no gore, only small amounts of blood, and when Koro-sensei is injured, his skin drips like slime to represent blood.
