S2 EP1 around the first quarter the Matsu's are shown as fatter versions of themselves. (excluding Ichimatsu) They were portrayed as more disgusting than when they were skinny. Iyami and Chibita call them "gross" after seeing them. This was meant to be interpreted as funny by the viewer.

It happens episode 14 of season 2. We don't see or hear the actual crash occur, but we're shown both an over view and close-up of the two vehicles that collided. Those vehicles are a bus and a small car. The driver of the small car is shown to be bleeding from what we can assume are several injuries, though we don't don't see the injuries themselves.

There are no canonically LGBT characters & the LGBT community is rarely even alluded to.

In the first instance, the character doesn't die. A girl nearly attempts suicide but is stopped by one of the brothers. ["Jyushimatsu Falls In Love"] In the second instance it is a joke, but the character technically dies. ["Osomatsu-san, Such As It Was"] In the third instance, the character does not die but the episode is very heavily based around suicide and it is in between being taken seriously and not. ["Iyami-san Is Troubled"]

In Osomatsu Kun (the manga) the chapter "The Terrifying Lodger" is about a character name Tougou who forces Osomatsu to commit bad actions such as stealing with him. If he was to tell his family Tougou threatened to kill him and would often be seen holding a knife to Osomatsu.

Not active misgendering but there are multiple jokes that have "men in dresses" as the punchline, usually perpetuating the stereotype that trans women are sexually predatory.

There is no eye mutilation, but there are two times when the Matsu's were poked in the eyes by fingers, Karamatsu's eyes and Osomatsu's eyes.

Not necessarily hate speech, but there is transphobia and occasional colorism.
