Papi is a controlling father who emotionally neglects his son and on multiple occasions, attempts (and sometimes succeeds) to physically 'discipline' him or even kill him. Kedamono is also implied to have been abused in his youth by his mother, the White Wolf.
Popee is shown to be unable to regulate his anger and often releases it on Kedamono. Paola, the elephant car, is an elephant whose head was implied to have been cut off to make the car functional (as cars can work via animal heads in this universe). The frog is wounded or killed multiple times.
Kedamono, the deuteragonist, dies dozens of times over the course of the entire show, as well as some deaths from Kaeru the frog and Onomadek, Kedamono's mirror (another wolf).
Popee turns into a fly in the Elephant episode before being swatted away by Paola. During the Karate Show episode, cockroaches stream out of a garbage can. In the Hypnosis episode, Popee and Papi end up hypnotizing one another and while their physical appearances look the same, they take on the behavioural traits of worms/caterpillars and are eaten by the other characters, hypnotized into behaving like birds. In the Medicine episode, Popee becomes deathly ill and falls unconscious, later having his face (and the soup left in his mouth) be surrounded by flies.
Not exactly, but in the episode "Poison" Popees father kisses him (to suck out poison) despite him clearly being uncomfortable and fighting against it.

this happens fairly often. Noteably in S1E2 "Limbo" when Kedamonos tail catches fire, S1E3 "Acrobatics" when Popee is struck by lightning and his charred body is shown, and S1E8 "Fire Ring" when literally all appearing characters catch fire at some point.
During the Pantomime episode, where Popee and Kedamono play with their imagination(s) and use imaginary items in the real world, Popee chokes (and hangs) while being pulled back into a cliff by a neck rope and eventually falls in with Kedamono.
Most notably, during the Knife Game episode, there's a visual gag where Popee pretends to have stabbed his finger, though he reveals that the knife went through the space between the five fingers in his glove instead.
In the 13th episode of season 1, "Dance", Popees eyes fall out and the rest of the episode is dedicated to replacing them with various objects. In S3E5 "Sleep" Popee rips off his eyelids.
Papi vomits blood before his death in the Poison episode and in the Dark Side episode, he also throws up after realizing he's been fed pieces of Paola's trunk.
Popee is incredibly self-destructive and a lot of his behaviours could easily be interpreted as forms of self harm. In addition to this fact, he and multiple other characters harm (or kill) themselves to get out of a variety of rough situations.
Popee is tied up in the Escape Show episode only to be unable to pull himself out of his bounds. He is terrified and begs his assistant for help while the time bomb ticks away. He is later strapped to a cot by the Alien in the same episode, and while he's able to free his arms, he doesn't make it out alive. Kedamono is also restrained in the Knife Thrower episode.
Popee attempts suicide and holds a gun to the side of his head in the Poison episode. Papi commits a murder-suicide in the Ghost episode in order to escape Popee's wrath. Papi bleeds to death after he inserts a katana into his nostril in the Eraser episode. Papi commits the ritual act of seppuku in the Opening episode, but nothing is shown and it is brief.
Wakako Masuda confirmed that Papi is capable of romantic attraction to any individual, regardless of gender, and he dies multiple times. Popee is implied to be bi-questioning in a tweet from one of the creators whilst having a unique gender identity and also dies multiple times.
Vs. God is about Popee and Kedamono meeting and messing with their universe’s god, only to get shot by their laser eyes. Christian iconography, often related to death, also appears.
The Face episode is highly depressing. As of me writing this, the majority vote says there's no sad ending and I'm honestly stumped as to why that is? Popee and Kedamono, after Kedamono's identity as a wolf is exposed by the frog (which completely shatters him), both drop to the floor in anguish, unsure of what's coming next but understanding they only really have each other for company.
