The Kuji brothers relationship is complicated, and could be construed as abusive. Chikai does grab Toi by the face once, but never again after that. While Chikai does make a point to ask Toi if he really wants to be involved with him, and regularly attempts to abandon him for his own good, he does allow Toi to be involved in his criminal activities, and is not really a "good" brother.

No animals are hit, but several cats are shaved for their fur, and one character does threaten to cut open a cat because the cat swallowed something it shouldn't have. Nothing graphic is ever shown, and this is all done for comedy purposes rather than horror.

Throughout the series, there is non-sexual, but non-consensual anal penetration for plot purposes, most of it played for laughs. If you are uncomfortable with anything in the first episode, you will probably be uncomfortable with the rest of the series, as the things shown are central to the plot.
In episodes 7, 9, and 10, we are shown more explicitly that a character is sexually involved with another, but it is not revealed that it is non-consensual until episode 10.
Additionally, all the "kappa-zombies" have some sort of paraphilia, and some are either nonconsensual or dubiously consensual, but none are shown graphically.

There are two scenes of waterboarding throughout the show, but they are sort of in the background of the scenes where they occur in, mostly underneath dialogue.

There is a crossdressing subplot that is resolved within the first episode. While a character makes an assumption about a character's gender based on the underwear that character wears, he is not incorrect, and the character continues to wear his costume without any further issues.

[Slight spoiler] A character is revealed to have ended up in their wheelchair after being hit by a car. It occurs in a flashback segment in episode 5, we don't see the impact on screen, but we hear it. We are then shown the character's legs while they're on the ground. They're battered, but there is no blood or gore.
