ReportSynopsis
Uta — the most beloved singer in the world. Her voice, which she sings with while concealing her true identity, has been described as “otherworldly.” She will appear in public for the first time at a live concert. As the venue fills with all kinds of Uta fans — excited pirates, the Navy watching closely, and the Straw Hats led by Luffy who simply came to enjoy her sonorous performance — the voice that the whole world has been waiting for is about to resound.

Fan favorite character "Uta" is suffering in the early stages of extreme psychosis when she makes comments in anger calling out fan favorite character "Red haired Shanks" (father figure for Uta, and role model for main character Strawhat Luffy) as extremely negligent by way of "sudden unexplained departure", "sudden and indefinite no contact", "displays of extreme violence".
It is revealed towards the end of the movie, while Uta is "near death", that she had been able to rewatch some security camera type of footage that was recorded during the night prior to Shank's sudden departure. Revealing that the "display of extreme violence was unexpectedly caused by her

Story trope; popular music idol struggles with depression while overdosing on stimulants. Self sacrifice by sleep deprivation using (magical) drug enhancement.
Major shift in tone (countdown, horror) when fan favorite character "Uta" is portrayed as suffering from severe depression, and her outward display of manic joy and goodwill are a manifestation of very *literal* psychosis due to Uta's prolonged abuse of (mushroom shaped, potent stimulant) uppers called "wake shrooms".
Intense discovery that Uta is enacting a planned (mass) suicide by method of a magically enhanced stimulant overdose causing death by overexertion from sleep deprivation.
Some backstory is provided only about Uta to explain that her addiction (to her fans, "making music", using wake shroom as performance enhancement) is a consequence of prolonged social isolation; combined with "workaholic" self sacrifice, ambition for the movie's climax of very literal (magical) depersonalization, intense real life & magical derealization, processing childhood shame over the sudden departure of a father figure, and intense fear of "the world government".
Early scene in movie depicting fan favorite character "Blackfoot Sanji" discovering the first wake shroom in the movie. He is picking through possible ingredients to cook for others, and without dialogue or much animation he appears to question why the wake shroom is available and throws it into the ocean. "Casual and regular" depictions of addiction to "drinking" (sake) and "smoking" (ambiguous, minor stimulant) are less frequently shown from characters but are not brought up into further discussions about addiction.

It’s not what I would count as drugs because it’s only effect is keeping a person awake and makes them aggressive/irritable from lack of sleep.

The entire cast is willing to sacrifice themselves to stop the antagonist but this doesn’t occur. The antagonist is trying to force everyone into a dream dimension she can control where there is no suffering. She is slowly killing herself to make this reality but she would still be alive in the other dimension. She comes to her senses and dies at the end of the movie to atone for her actions.

I would say it’s debatable. A character dies from eating mushrooms intentionally, but when she started she knew she would still be alive in an alternative utopian reality. She closes the reality and get others out of it and back to their original lives.
