One of the ghosts is perpetually high on hallucinogens, this doesn't bother her and is played for laughs. A different ghost died of an overdose/bad combination of drugs at a party - there's a flashback scene showing the party, everyone is high on cocaine and/or unknown pills. Yet another ghost extols the virtues of cocaine and laudanum, which she used liberally when she was alive.

A Victorian ghost mentions cocaine a fair few times, and says that when she was alive she took laudanum (alcohol + morphine) in order to sleep. It's not definitively addiction, but it could certainly be interpreted that way.

In s1e04, a neighbor gets drunk on wine at dinner.
In s1e16, flashbacks show how one of the ghosts died, at a party with drugs and booze. The friends he was with had a prank/game that involved basically tricking someone into drinking, resulting in the classic "chug, chug, chug, chug!" chanting.

If I remember correctly, there has only been one death of a living character that took place during the events of the show. It occurs in season 3, and it’s a recurring but fairly minor character. Some of the deaths of ghosts are also shown, and ghosts getting sent to heaven/hell is treated similarly to death.

One of the main ghosts was a father. s1e06 ("Pete's Wife") centers around his family, though it focuses on his perspective. His daughter is briefly shown, it's a positive moment. Also, in s1e11 ("Sam's Mom"), we learn that Sam's mother is dead, and we see Sam is grieving still/again.

No, however the firm Trevor worked for is a real company whose offices were in the World Trade Centre, so there is a possibility (though probably unlikely in a sitcom) that it could be mentioned in future episodes that his co-workers would have been victims/survivors of 9/11.

In the very first episode, Sam (main character) has an accident and goes to the hospital. She is carried out on a gurney, has a CT scan done, and is shown unconscious beside a monitor. The show momentarily implies she has died (misleading dialogue between husband and doctor), but we're immediately shown that she is currently alive and well. The entire sequence lasts ~1-2 minutes.
