Not depicted as a joke, but: Through out several episodes several men are asked (a.k.a. told) to have sex with the/both female shogun(s). In one episode a monk is forced to have sex with a female prostitute to gain sexual experience before 'bedding' the female shogun, when he first refuses another monk and an other prostitute are killed.

The first female shogun's (before it is made public the she was the only heir to the house of Tokugawa) sole purpose is to produce a male heir (and not through marriage). Until she has succeeded this task, her deceased father's wet nurse who later acted as his right hand is hell bent on keeping the identity and gender of the girl/woman a secret.
