In Performance Review (2x08), brief mention is made of a past co-worker who shot himself due to depression. While other characters do take the matter seriously, Michael brushes it off by saying, Oh yeah, that guy was weird." In addition to this, one of Dunder Mifflin's founders (Mifflin) also shot himself, a fact which is mentioned throughout the show. Off the top of my head, it's definitely mentioned in Dunder Mifflin Infinity and Company Picnic. In Company Picnic, it's referenced in Michael and Holly's parody of Slumdog Millionaire -- one of the questions asks how Mifflin killed himself.

Yes. Ryan is largely implied to be addicted to cocaine after he becomes Michael's boss, and later tells Michael that he helped him get off drugs. Also, in the episode "Drug Testing" a half-smoked joint is found in the parking lot. Nobody is shown smoking it.

There is also some sexual harassment: In an earlier season Pam states that sexual harassment awareness day is the day people usually like to sexually harass her as a joke. Michael then tries to get her to make out with a blow up doll as a joke later in the episode.
