It doesn't go anywhere, especially with Predaking, but it can absolutely be read as the start of something abusive with Knock Out, most obviously in the episode Thirst. I'm not fully certain it qualifies, but it's better to be safe than sorry. Ratchet drugs himself with synth-en, Megatron and Starscream drug both themselves and multiple corpses with Dark Energon, and in S3 it's revealed that Knock Out has been testing new synth-en formulas on an alive prisoner. Nobody is drugged into unconsciousness though, even if Ratchet does pass out after the first synth-en dose.
