Ms. Fortune (a cat-person) does, non-canonically, die in one Story Mode ending.

Beowulf wears a wolf skin as a hood, but its death is not shown as it is implied to be something he has had for a long time

All story modes require the death of the current Skullgirl, Marie, in order to finish. Marie is around Peacock's age, making her about 12-15. Squigly also died at 14 years old, although she resurrects as a zombie every time the Skull Heart appears.

Many characters are noted to be orphans or with a dead parent/parent figure (Peacock, Marie, Cerebella, Parasoul, Ms. Fortune, etc). Squigly's father, Roberto, and (on the mobile app) Parasoul's mother Nancy, are the only times the deaths are shown on screen, though.

One of the playable characters, Valentine, has a move in which she readies a syringe, which is required to then throw said syringe at the opponent.
