At the end of the game, a character sacrificed himself along with his fiance and got married. He didn’t get to say goodbye to his closest associate due to her falling unconscious. She can be seen breaking down and crying when she wakes up, and still grieving his loss if she is talked to again.

Debatable. A character's parents disapprove of him dating another character (think Romeo and Juliet), and they put a curse on his lover to force them to be apart. In chapter 7 two people create a child destined to die and lies to her, telling her that she is their daughter. She feels betrayed, but if you return to that location after the game you find out that she survived and is happily living with her family.

There are various enemies who can be killed for points, but none are particularly animal-like, more monster like. Some do resemble pigs or mammoths, however.

Like with any Mario, you jump on enemies to progress through the levels. The game introduces other ways of doing this, including a Pixl that is a bomb and another that is a hammer. Some enemies are meant to look like IRL animals, including pigs and turtles among others.

There is a joke scene when you first enter World 4, which is just straight up space, where you have the option to say no to using the space helmet. Saying no too many times results in a game over but it's mostly played for laughs.

The main villain's plan is intended to bring about his own death as much as it is the death of other people. The game uses "end [their] games" to describe in-universe deaths to obfuscate their nature, but the character's despair is made explicit.

While not exactly Hell, a later location in the game is called the Underwhere, with various nods to the Underworld and Hell throughout. There are demon-like NPCs in the Underwhere.
An answer a character gives on a game show minigame earlier in the game references demons as well.
